Diapositive1Notre équipe a été initialement créé en 2011 (équipe 3 de CarMeN: Glucolipotoxicité, Stress métabolique et Diabètes). Elle implique des chercheurs fondamentaux et des cliniciens avec des compétences complémentaires dans le domaine de l’action et de la sécrétion d’insuline. Notre objectif était d’identifier les mécanismes moléculaires communs par lesquels le stress métabolique associé à la glucolipotoxicité affectait l’action de l’insuline dans le foie et le muscle et la sécrétion d’insuline par les cellules béta du pancréas, contribuant ainsi aux deux composantes du diabète de type 2 (DT2). Au cours des dernières années, nous nous sommes focalisés sur l’implication des mitochondries et du réticulum endoplasmique (RE) dans les altérations fonctionnelles de ces tissus, mettant en évidence pour la première fois un rôle clé des interactions structurelles et fonctionnelles entre ces deux organites dans les altérations de l’action et de la sécrétion de l’insuline au cours du DT2. Par conséquent, notre équipe a évolué et se nomme maintenant « Communication inter-organite et diabète » (Orcadia, 2016-2020). Notre objectif est de clarifier les mécanismes par lesquels les interactions mitochondries-RE sont impliquées dans la pathogenèse du DT2 et si le contrôle de ces interactions pourrait constituer une nouvelle cible pour améliorer l’action et la sécrétion d’insuline dans le DT2 (Figure 1).

La communication entre les organites intracellulaires permet à la cellule d’adapter son métabolisme en fonction de son environnement, afin de maintenir l’homéostasie cellulaire. Plus précisément, le RE et les mitochondries interagissent au niveau de points de contact, appelés MAMs (mitochondria-associated membranes), afin d’échanger des lipides et du calcium, 2 molécules signal qui jouent un rôle clé dans l’homéostasie métabolique. En effet, la signalisation lipidique et calcique contrôle à la fois l’action et la sécrétion d’insuline, expliquant pourquoi ces deux organites jouent un rôle important dans la pathogenèse du diabète de type 2 (DT2).

Nous avons récemment démontré un nouveau rôle des interactions mitochondries-RE dans le contrôle de l’action de l’insuline dans le foie et le muscle squelettique, ainsi qu’un rôle d’une altération des MAMs dans le développement de l’insulino-résistance hépatique et musculaire (Tubbs E et al. Diabetes 2014, Rieusset J et al. Diabetologia 2016, Tubbs E et al. Diabetes 2018) (Figure 2). Un défaut des MAMs est également associé à des altérations de la sécrétion d’insuline dans les cellules bêta du pancréas (Thivolet et al. PLoS One 2017). Par conséquent, l’ensemble de ces données met en évidence un nouveau mécanisme, commun à l’insulino-résistance périphérique et aux défauts fonctionnels des cellules béta, dévoilant de nouvelles stratégies préventives et/ou thérapeutiques pour les deux composantes du DT2 (Rieusset J Adv Exp Med Biol 2017).

En parallèle, nous avons analysé la régulation dynamique des MAMs en situation physiologique et pathologique, afin d’identifier de nouvelles cibles intracellulaires pour moduler les interactions ER-mitochondries, et potentiellement améliorer l’homéostasie du glucose. Nous avons récemment démontré que les interactions mitochondries-RE sont régulées au cours des transitions alimentaires, et plus particulièrement elles sont réduites en postprandial, plus particulièrement en présence de concentrations élevées de glucose, afin de diminuer la dynamique et la fonction des mitochondries et orienter les flux de glucose vers le stockage (Theurey P et al. J Mol Cell Biol 2016) (Figure 3). De manière intéressante, cette régulation nutritionnelle des MAMs est perdue dans le foie des souris obèses et diabétiques. Par conséquent, les MAMs pourraient constituer une nouvelle plate-forme de détection du glucose qui permettrait aux hépatocytes d’adapter leur métabolisme aux transitions nutritionnelles, et la perte de cette régulation pourrait participer à l’inflexibilité métabolique associée à l’obésité et au DT2 (Theurey P et al. Trends Endocrinol Metab 2017).

L’objectif général de notre recherche est donc de mieux caractériser la nature et l’importance physiologique des MAMs dans l’homéostasie du glucose et leur rôle dans la pathogenèse du DT2. Pour cela, nous combinons des analyses dans des modèles cellulaires, des modèles murins de DT2 et chez l’homme. Plus précisément, nos objectifs spécifiques sont :

  • D’identifier la nature moléculaire des acteurs des MAMs et leurs rôles fonctionnels,
  • De caractériser la signification physiologique des MAMs dans le contrôle de l’action et de la sécrétion d’insuline,
  • D’identifier les régulateurs des MAMs et leurs impacts fonctionnels,
  • De valider si les MAMs pourrait être une nouvelle cible pour le traitement du DT2.

Globalement, la combinaison d’approches cellulaires et intégrées, tant chez les animaux que chez l’homme, couplée à des outils d’imagerie innovantes, devrait nous permettre de clarifier le rôle de la communication mitochondrie-RE dans la physiopathologie du DT2 et d’ouvrir de nouvelles voies pour le traitement de cette pathologie.

    • Analyse structurale et fonctionnelle des interactions mitochondrie-RE. En particulier, nous avons développé et validé une nouvelle méthode pour détecter et quantifier les interactions ER-mitochondries par in situ Proximity ligation assay (PLA), via la visualisation des interactions VDAC1-IP3R1, Grp75-IP3R1 ou CypD-IP3R1 dans des cellules fixées ou des tissus enrobés de paraffine (Figure 4).

     

    • Nous analysons également les interactions mitochondrie-RE par fractionnement subcellulaire, par microscopie électronique et par co-immunoprécipitation, et nous mesurons le transfert de calcium du RE aux mitochondries par microscopie confocale en utilisant des sondes Caméléon.
